About Acetic Water

Acetic water is water containing a small amount of acetic acid (CH3COOH, the weak organic acid that gives vinegar its taste).[1] The result is a slightly acidic liquid used in research laboratories as a diluent for lyophilized peptides that do not dissolve well in neutral water. Pepitiva supplies acetic water in 3 mL and 10 mL vials.

What is in it?

  • Water
  • Acetic acid (formula C2H4O2, molecular weight 60.05 g/mol)[1] at a low concentration. The exact concentration of each lot is stated on the vial label and the Certificate of Analysis.

Acetic water contains no preservative. For a multi-use diluent with a bacteriostatic preservative, see bacteriostatic water.

For research and laboratory use only. Not for human consumption.

Mechanism of Action

Why an acidic diluent helps some peptides

A peptide's solubility depends on its net charge, which changes with pH. Peptides rich in basic residues (lysine, arginine, histidine) carry more positive charge at acidic pH, which tends to keep the molecules apart in solution instead of clumping together. For such peptides a dilute acid can dissolve material that stays cloudy in plain water. Peptides that are acidic or neutral do not necessarily benefit, so the diluent should follow the documentation for each compound.

Research Applications

Use in the laboratory

  • Reconstituting lyophilized peptides whose documentation calls for a dilute acid diluent.
  • Preparing stock solutions that are later diluted into buffers for in vitro assays.

Handling and storage

Keep the vial sealed and use aseptic technique when withdrawing liquid. Do not use the solution if it is cloudy or the seal is damaged.

Frequently Asked Questions

What is acetic water?

Water with a small amount of acetic acid added, giving a slightly acidic diluent for laboratory use with peptides that dissolve poorly in neutral water.

What is the difference between acetic water and bacteriostatic water?

Acetic water is slightly acidic and has no preservative. Bacteriostatic water is close to neutral and contains 0.9% benzyl alcohol as a preservative. See bacteriostatic water.

What concentration of acetic acid does it contain?

The concentration of each lot is printed on the vial label and in the Certificate of Analysis.

When is acetic water used instead of bacteriostatic water?

When a peptide's documentation specifies an acidic diluent, typically for peptides rich in basic amino acids that stay cloudy in neutral water.
